Allerdings hatten diese Technologien bisher einen relativ geringen Einfluss auf die Entwicklung von medizinischen Implantaten oder die Verbesserung von klinischen Behandlungsmethoden.\r\nDer Einsatz von bildgebenden Verfahren zur Extraktion von wichtigen funktionellen Informationen und der Kopplung von Bildinformationen mit physiologischen Aspekten, die nicht durch Bildgebung allein verstanden werden können, stellt eine große Herausforderung dar. In silico Modelle, also computerunterstützte Simulationen, bergen enormes Potenzial zur Bewältigung dieser Herausforderungen und können zu einem besseren Verständnis der Herzfunktion, sei es im gesunden oder auch im erkrankten Zustand, führen. Ein entscheidender Vorteil ist hierbei die Möglichkeit diagnostische und therapeutische Methoden zu optimieren ohne dabei Patienten zusätzlichen Risiken oder Schmerzen auszusetzen. In silico Experimente können somit bei der Untersuchung von klinisch relevanten Problemen, wie zum Beispiel Herzklappenchirurgie, Ballon-Angioplastie oder Stenting, äußerst hilfreich sein.\r\nDie Herzfunktion ergibt sich aus der Interaktion verschiedener Physiken, unter anderem der Elektrophysiologie, der Strukturmechanik und der Strömungsmechanik. Während die beteiligten Physiken klar bidirektional wechselwirken, werden sie in praktisch allen Studien isoliert betrachtet. Dies ist zu einem guten Teil auf die Komplexität von Multiphysik-Modellen und den Schwierigkeiten, die bei der Entwicklung von modernen numerischen Methoden und deren effizienten Implementierung auf modernen Rechnerarchitekturen auftauchen, zurückzuführen. Die Bewältigung dieser Herausforderungen ist das zentrale Themas dieses Projektes und die Durchführung setzt auf die Kombination der Expertisen der Antragsteller: Christoph Augustin und Gernot Plank in der Modellierung der Elektrophysiologie und der Strukturmechanik und Shawn Shadden in der Computermodellierung des Blutflusses.\r\nDieses ambitionierte Unterfangen geht weit über den aktuellen Stand der Technik hinaus. Kombinierte Modelle von Herz und Blutgefäßen ermöglichen die Berechnung von natürlichen Randbedingungen für die Simulation des Blutflusses und das Studium von Rückkopplungsmechanismen die die Pumpleistung des Herzens beeinflussen können. Im Endeffekt wird dieses Modell eines der modernsten Werkzeuge der Grundlagenforschung sein, das einen mechanistischen Einblick in die Herzpumpfunktion gewährt. Von wesentlicher Bedeutung ist in diesem Kontext auch die Überprüfung, Parametrisierung und Validierung dieses Modells mit klinischen Datensätzen, die von klinischen Partnern zur Verfügung gestellt werden.\r\nDas Ziel dieser Forschung ist ein innovatives Multiphysik-Computermodell der gesamten Herzfunktion um eine biophysikalisch detaillierte und bidirektional gekoppelte Erforschung der elektromechanischen Funktion des Herzens im gesunden wie im kranken Zustand zu ermöglichen.", "en": "Advances in tomographic imaging have enabled unprecedented ability to image cardiac anatomy. However, these technologies have had relatively modest clinical impact. Key challenge have been the use of imaging to extract important functional information and to understand how image-derived information couples with other physiological aspects that cannot be understood through imaging alone. In-silico models hold vast potential to address these challenges by enabling the integration of imaging data into quantitative frameworks that can aid in better understanding cardiac function in health and disease, and developing optimal treatment strategies.\r\nCardiac function arises from interactions between different physics, including electrophysiology, structural mechanics and fluid dynamics. While these physics are bidirectionally linked, virtually all reported modeling studies have considered them in isolation. In no small part this is due to the complexity of multiphysics models and the difficulties in devising advanced numerical methods and their efficient implementation on modern computing architectures. Addressing these challenges as we propose is key to keep model execution and analysis cycles tractable and, thus, to leverage modeling in a translational context. The proposed work goes beyond the state-of-the-art in many regards. Combined models of both heart and attached outflow vessels will be used to provide natural inflow boundary conditions for simulating blood flow and to facilitate studying feedback mechanisms between flow and pressure in the outflow vessels and pumping performance. This set of novel features and the parallel efficiency of the implemented methods will provide a unique platform for translational research. At the fundamental level, this model will be among the most advanced basic research tools for gaining mechanistic insight into cardiac pumping function. This challenging endeavor is feasible only as it builds upon combining the expertise of the applicant Christoph Augustin (Medical University of Graz) in modeling electrophysiology and soft tissue mechanics, and the host Shawn Shadden (UC Berkeley) in computational modeling of blood flow. Clinical datasets for model parametrization and validation are provided by Titus Kühne at the German Heart Centre Berlin, as well as by clinical collaborators of Prof. Shadden at the University of California San Francisco.Technical challenges to be addressed relate to multiphysics coupling methods which converge robustly and scale efficiently. While such methods have been developed recently for single physics cardiac simulations, this has not been the case for coupled multiphysics models of total heart function. This will be addressed by massively parallel implementations which are based on domain decomposition and strongly scalable iterative methods using appropriate preconditioning and stabilization techniques. In summary, the overall objective is to develop the most advanced in-silico model of total heart function, to facilitate biophysically detailed studies of electro-mechano-fluidic function of the heart. Members of the human TRP Canonical (TRPC) subfamily\r\n(TRPC1/3/4/5/6/7), form channels that mediate Ca2+- and Na+-ion entry and are\r\nexpressed in many mammalian cell types to serve a broad-range of important\r\nphysiological processes. Recently, TRPC3/6 channels have been implicated in the\r\npathogenesis of cardiovascular disorders including heart failure, arrhythmias, and\r\nsudden death. Consequently, TRPC channels have emerged as highly promising\r\ntargets for drug development, while, at the same time, their exact cellular function and\r\nprinciples of pharmacologic modulation are still incompletely understood and require\r\nbetter understanding. For this, novel strategies to experimentally control channel\r\nfunction are urgently needed. These channels are endogenously controlled by plasma\r\nmembrane lipid metabolites, which are difficult to administer in most experimental\r\nsettings. Novel modulating or blocking ligands with potentially suitable selectivity for\r\nexact experimental control of TRPC3/6 channel have just recently been reported. One\r\nobjective of this proposal is therefore the synthesis of small modulators\r\n(inhibitors/activators) of TRPC3/6-channels that response to light, allowing the\r\ncontrol of biological events with unparalleled spatial and temporal precision – so\r\ncalled “photoswitches” (e.g. azobenzens). Taking advantage of our experience in the\r\nsynthesis of pyrazole-based inhibitors of TRPC3/6-channels and benzimidazolonebased\r\nactivators as well as our expertise in microwave and continuous-flow\r\nchemistry, we aim to generate and explore various photoswitchable ligands in\r\nconnection to the emerging pharmacology of TRPC3/6-channels. Another objective\r\nof ours, involving small photoresponsive molecules, is the synthesis of photoaffinity\r\nlabels to help in the elucidation of potential ligand-binding sites in the TRPC3/6-\r\nchannels as well as on the structure elucidation of TRPC3/6-channels.\r\nThe research proposed herein is meant to become a significant contribution to\r\na thrilling and rapidly growing field for optoregulation of biological functions in\r\nmammalian cells and for better understanding on the TRPC3/6-channel machinery.
