Die ESA handelt im Namen ihrer Mitgliedstaaten, die alle Vertragsparteien des Weltraumvertrags sind. Als solche müssen die Aktivitäten der ESA mit den Verpflichtungen der Mitgliedstaaten gemäß Artikel IX übereinstimmen. Die derzeitigen PP-Standards, die auf den Methoden der Viking-Mission der NASA aus den 1970er Jahren beruhen, sind veraltet. Unser Verständnis von mikrobiellem Leben hat sich weiterentwickelt, und so muss auch unser Ansatz für PP. Wir schlagen ein revolutionäres Projekt im Einklang mit der PP-Vision der ESA für 2026 vor (ESA-TECQI-TN-2024-000152). Unser Ziel ist es, den Standardtest (ECSS-Q-ST-70-55) mit einem hochmodernen probabilistischen Risikobewertungsmodell zu erweitern. Jüngste Forschungsarbeiten (Green et al., 2023) haben mehrere Wissenslücken aufgedeckt, wie z. B. die Überlebensfähigkeit von PP-relevanten Mikroorganismen, die wir nun systematisch angehen wollen. Zu diesem Zweck werden wir modernste Metagenomik als Schlüsseltechnologie zur Bewertung der Biodiversität und des Risikos von Vorwärts- und Rückwärtskontaminationen einsetzen. Durch die Integration von Echtzeit-Sequenzierungstechnologien wie Nanopore können wir zeitnah (<24h) ein Profil von Umgebungen mit geringer Biomasse erstellen und den Ingenieuren ermöglichen, Maßnahmen zu ergreifen. Um die Anpassung von Mikroben an Reinraum- und Raumfahrzeugumgebungen wirklich zu verstehen, werden wir Metabolitenvorhersagen auf Genomebene und Massenspektrometrie von Proteinen und Metaboliten durchführen. Wir planen die Entwicklung eines überwachten maschinellen Lernmodells, das metagenomische und massenspektrometrische Daten nutzt, um einen risikoinformierten Wert für die Überlebensfähigkeit von Mikroben in Reinräumen und auf Oberflächen von Raumfahrzeugen zu ermitteln.                 "en": "Article IX of the Outer Space Treaty (OST) (https://www.unoosa.org/oosa/en/ourwork/spacelaw/treaties/outerspacetreaty.html) requires that State parties adopt appropriate measures to avoid harmful contamination of celestial bodies and adverse changes on Earth. ESA acts on behalf of its Member States, all of which are State parties to the OST. As such, ESA's activities shall comply with Member States' obligations to Article IX. The current PP standards, rooted in methodologies from the 1970s NASA Viking mission, are outdated. Our understanding of microbial life has evolved, and so must our approach to PP. We propose a revolutionary project in line with ESA’s PP vision for 2026 (ESA-TECQI-TN-2024-000152). Our goal is to enhance the standard assay (ECSS-Q-ST-70-55) with a cutting-edge probabilistic risk assessment model. Recent research (Green et al., 2023) has unveiled several knowledge gaps, such as the survivability of PP-relevant microorganisms, which we aim to address now systematically. For this, we will leverage state-of-the-art metagenomics as a key technology to assess biodiversity and the risk of forward and backward contaminations. Myelin acts as the electrical insulation of neuronal fibers andis essential for motor, sensory, and cognitive functions and is composed of several lipids and proteins arrangedin a specific geometry. However, myelin loss occurs regionally in inflammatory diseases like multiplesclerosis, myelitis, and optic neuritis, and it also serves as a biomarker of aging.Despite the availability of various MRI-based approaches for detecting myelin, there remains a significant gapin systematically validated knowledge regarding how myelin's lipids, proteins, and structural variationsinfluence MRI signal generation. Furthermore, there is no gold standard for assessing the constituents ofmyelin, and conclusions are primarily based on MRI of formalin-fixed tissue.Quantitative post-mortem MRI of unfixed human brains in situ, combined with subsequent mass spectrometryimaging (LA-ICP-TOF-MS, MALDI-MSI), small-angle X-ray scattering (SAXS), and microscopy (CARS,TEM, IHC), will enable histologically enriched modelling of tissue and the simulation of fundamental MRIparameters (relaxometry, susceptibility, and diffusion) to reveal causal relationships with chemical elements,matrix composition, structure, and common molecules of myelin, neural fiber orientation, and biometals. Inaddition to 20 control brains, 10 brains from deceased individuals with multiple sclerosis will be recruited tobroaden the spectrum of myelination under investigation.This research addresses the fundamental biophysical mechanisms underlying MRI of myelin and chemicalelements, which currently lack validation. We will establish a publicly available elemental and histochemicalatlas of the human brain and identify how histological features causally contribute to quantitative MRIparameters.",
