                "en": "Human placental villous trees are covered by a two-layered epithelium-like trophoblast layer, which consists of mononucleated cytotrophoblasts and an overlying multinucleated syncytiotrophoblast. Cell turnover in the trophoblast layer includes proliferation, differentiation and subsequent fusion of cytotrophoblasts with the syncytiotrophoblast. Thus, both layers represent stages of different degree of differentiation, ranging from very early differentiated (cytotrophoblast) to terminally differentiated stages (syncytiotrophoblast). Data on comparative whole transcriptome analyses between in vitro cultured mononucleated and multinucleated primary trophoblasts exist, but have their clear limitations. Withdrawal from the in vivo microenvironment as well as in vitro culture of primary cells alters gene expression profiles, which no longer resemble the in vivo situation. To overcome these limitations we intent to microdissect these two cell fractions directly from fixed, paraffin embedded placental tissue. Total RNAs from microdissected cytotrophoblasts and syncytiotrophoblast will be subjected to microarray analysis with the aim to detect differentially expressed genes. The expectations in this project are to gain insight into the spatial expression of genes relevant for trophoblast differentiation. Furthermore, this project should deliver the basis for follow-up studies comparing placentas from healthy and pathologic pregnancies."

                "de": "Genetische Pränataldiagostik erfordert die Gewinnung von Zellen des Kindes. Dies erfolgt durch invasive Methoden wie Amniozentese oder Chorionzottenbiopsie. Das Risiko, daß es durch diese Eingriffe zu einem Abort kommt, ist nicht vernachlässigbar und liegt bei 0,5 – 1%.  Aus diesem Grund ist genetische Pränataldiagnostik auf Schwangerschaften mit einem erhöhten Risiko einer genetischen Krankheit des Fetus beschränkt, wie z. B. einem Alter der Schwangeren von über 35 Jahren. Tatsächlich wird aber die Mehrzahl von Kindern mit genetischen Krankheiten nach Schwangerschaften geboren, die nicht primär als mit erhöhtem Risiko behaftet eingestuft werden können und bei denen das Risiko einer invasive Diagnostik in keinem vertretbaren Verhältnis zum Risiko durch den Eingriff steht.\r\nAusgehend von einer von uns entwickelten Methode zur geschlechtsunabhängigen Analyse der fötalen Identität von Zellen wollen wir in dem vorgeschlagenen Projekt ein Verfahren etablieren, das auf der Basis der Analyse von Einzelzellen eine nicht-invasive pränatale Diagnostik numerischer Chromosomenaberrationen und monogenetisch bedingter Krankheiten ermöglicht. Wir vergleichen die Effizienz etablierter Protokolle zur Anreicherung fetaler Zellen aus dem peripheren Blut schwangerer Frauen. Weiters etablieren wir die Amplifikation des gesamten Genoms von Einzelzellen, kombinieren die Analyse der fetalen Identität mittels array comparative genomic hybridization zur Analyse von Trisomien und analysieren als Beispiel der prinzipiellen Machbarkeit Mutationen des CFTR Gens.\r\nAuf Basis dieser Arbeiten wollen wir unter Nutzung von Ressourcen der Medizinischen Universität Graz im Sinn einer kommerziellen Anwendung ein überregionales Zentrum zur zellbasierten nichtinvasiven Pränataldiagnostik etablieren.\r\n",
                "en": "Genetic prenatal diagnosis is dependent on procurement of fetal cells. This is done my invasive methods such as amniocentesis and chorionic villous sampling. These methods are associated to a risk of 0.5 – 1% of subsequent abortus. For this reason genetic prenatal diagnosis is restricted to pregnancies with are associated with an increased risk of renetik disease of the fetus, such as age of the pregnant women higher than 35 years. In fact the majority of children with genetic diseases is born at the end of pregnancies which have not abeen associated with an increased risk, where the risk of invasive procedures cannot be  justified.\r\nBased on a method which we developed previously for sex-independent analysis of the fetal identity of cells in a fetal microchimeric setting, we plan to establish a technique allowing for non-invasive prenatal diagnosis of both numeric chromosomal aberrations and monogenetic hereditary diseases, using analysis of single cells. We will compare the efficiency of established protocols for enrichment of fetal cells from the peripheral blood of pregnant women. Furthermore, we will establish whole genome amplification of single cells and combine the analysis of fetal identity with analysis of trisomy by means of array comparative genomic hybridization. As a proof of principle for feasibility of analysis of monogenic hereditary diseases using the preamplified genome of single cells we will analyze mutations of the CFTR gene.\r\nThe results of this work will be commercially exploited as we plan to establish an supra-regional center for cell-based non-invasive prenatal diagnosis by using the facilities of the Medical University of Graz.\r\n"

Zur Routinediagnostik einer Insektengiftallergie gehören Hauttests sowie die Bestimmung spezifischer, gegen Bienen- und Wespengift gerichtete Immunglobulin (Ig) E-Antikörper im Blutserum. Diese etablierten Diagnostikverfahren sind zwar relativ einfach durchführbar, führen aber häufig zu falsch-positiven Ergebnissen. In solchen Fällen hat sich in den letzten Jahren der Basophilenaktivierungstest basierend auf CD63 und CD20c aufgrund seiner hohen Spezifität und Sensitivität als sehr nützliche zusätzliche Methode erwiesen, ist in seiner gegenwärtigen Form jedoch eher ein qualitativer Test.\r\nZiel des Projektes ist es einen auf der Expression des Oberflächenmarkers CD11b-basierenden Basophilenaktivierungstest für die Diagnostik der Bienen- und Wespengiftallergie zu entwickeln und mit den Ergebnissen von CD63- und CD203c-basierenden Tests, sowie Standard-Diagnostikmethoden zu vergleichen.                 "de": "Uterussarkome sind mit 2-5% aller Uterustumoren als die seltensten Tumorentitäten des weiblichen Genitaltraktes zu betrachten. Innerhalb dieser Gruppe entfallen weniger als 15% auf endometriale Stromasarkome (ESS), weshalb die Pathogenese dieser mesenchymalen Neoplasien bisher sehr schlecht verstanden  ist.\r\n\r\nEndometriale Stromasarkome zeigen ein invasives Tumorwachstum in das umliegende Myometrium. Die betroffenen Patientinnen sind meist zwischen 42-58 Jahre alt. Die Tumoren treten oft prämenopausal auf. Es kommt sehr häufig zur Rezidivbildung, sowohl im Uterus, als auch in anderen Organen (Lunge, Knochen usw.). Bei sogenannten „low-grade“ endometrialen Stromasarkomen liegt die Rezidivrate in den ersten 10 postoperativen Jahren bei etwa 30-50%, bei „high-grade“ endometrialen Stromasarkomen kommt es oft, bereits in den ersten 2 Jahren nach Diagnose des primären Tumors, zur Rezidivbildung. Gegenwärtig ist die einzige Behandlungsoption eine operative Entfernung des Tumors bzw. der Absiedelungen in den betroffenen Organe (Uterus, Eierstöcke mit Eileitern usw.). \r\n\r\nChemotherapie, Hormontherapie und Strahlentherapie haben bisher keine befriedigenden Ergebnisse gezeigt. \r\n\r\nWir wollen die molekularbiologischen Grundlagen dieser seltenen Tumoren analysieren und verstehen. Es werden im Rahmen dieses Projektes morphologische und unterschiedliche molekularbiologisch Methoden anwenden.  \r\n",
