Als solche müssen die Aktivitäten der ESA mit den Verpflichtungen der Mitgliedstaaten gemäß Artikel IX übereinstimmen. Die derzeitigen PP-Standards, die auf den Methoden der Viking-Mission der NASA aus den 1970er Jahren beruhen, sind veraltet. Unser Verständnis von mikrobiellem Leben hat sich weiterentwickelt, und so muss auch unser Ansatz für PP. Wir schlagen ein revolutionäres Projekt im Einklang mit der PP-Vision der ESA für 2026 vor (ESA-TECQI-TN-2024-000152). Unser Ziel ist es, den Standardtest (ECSS-Q-ST-70-55) mit einem hochmodernen probabilistischen Risikobewertungsmodell zu erweitern. Jüngste Forschungsarbeiten (Green et al., 2023) haben mehrere Wissenslücken aufgedeckt, wie z. B. die Überlebensfähigkeit von PP-relevanten Mikroorganismen, die wir nun systematisch angehen wollen. Zu diesem Zweck werden wir modernste Metagenomik als Schlüsseltechnologie zur Bewertung der Biodiversität und des Risikos von Vorwärts- und Rückwärtskontaminationen einsetzen. Durch die Integration von Echtzeit-Sequenzierungstechnologien wie Nanopore können wir zeitnah (<24h) ein Profil von Umgebungen mit geringer Biomasse erstellen und den Ingenieuren ermöglichen, Maßnahmen zu ergreifen. Um die Anpassung von Mikroben an Reinraum- und Raumfahrzeugumgebungen wirklich zu verstehen, werden wir Metabolitenvorhersagen auf Genomebene und Massenspektrometrie von Proteinen und Metaboliten durchführen. Wir planen die Entwicklung eines überwachten maschinellen Lernmodells, das metagenomische und massenspektrometrische Daten nutzt, um einen risikoinformierten Wert für die Überlebensfähigkeit von Mikroben in Reinräumen und auf Oberflächen von Raumfahrzeugen zu ermitteln. ESA acts on behalf of its Member States, all of which are State parties to the OST. As such, ESA's activities shall comply with Member States' obligations to Article IX. The current PP standards, rooted in methodologies from the 1970s NASA Viking mission, are outdated. Our understanding of microbial life has evolved, and so must our approach to PP. We propose a revolutionary project in line with ESA’s PP vision for 2026 (ESA-TECQI-TN-2024-000152). Our goal is to enhance the standard assay (ECSS-Q-ST-70-55) with a cutting-edge probabilistic risk assessment model. For this, we will leverage state-of-the-art metagenomics as a key technology to assess biodiversity and the risk of forward and backward contaminations. By integrating real-time sequencing technologies like Nanopore, we can profile low biomass environments in a timely manner (<24h) and allow engineers to take action. To truly understand the adaptation of microbes to cleanroom and spacecraft environments, we will implement genome-scale metabolite predictions and mass spectrometry of proteins and metabolites. We plan to develop a supervised machine learning model that uses metagenomic and mass spectrometry data to provide a risk-informed value for the survivability of microbes from cleanrooms and spacecraft surfaces. Our innovative approach will modernize PP toolkits, provide more flexibility and increased efficiency to ESA mission teams when implementing PP inside industrial organizations while fulfilling ESA’s corporate obligations to conduct responsible and sustainable space explorations.
