                "de": "Rett Syndrom, eine schwerwiegende neurologische Erkrankung, durch de novo-Mutationen des MECP2 Gens verursacht, hat einen vierphasigen Verlauf: Nach einem mehr oder weiniger unauffälligen ersten Lebensjahr (Phase 1) kommt es zur Regression (Phase 2), in der erworbene Fähigkeiten wie Sprache und motorische Funktionen wieder verloren gehen. In dieser Zeit treten auch die für das Rett-Syndrom charakteristischen Handstereotypien auf. Mit Phase 3 tritt eine relative Stabilisierung des Entwicklungsstandes ein; Phase 4 ist gekennzeichnet von erneuten motorischen und kognitiven Beeinträchtigungen. Da eine Diagnose meist erst im Vorschul- oder Schulalter erfolgt, gibt es wenig systematisches Wissen über die Prä-Regressionsphase. \r\nZiel des eingereichten Projekts ist die Erstellung spezifischer Entwicklungsprofile dieser ersten Phase und zwar in Abhängigkeit von Locus und Typ (missense/nonsense) der Mutation. Unsere detaillierten Analysen der Frühzeichen sowie der Genotyp-Phänotyp Beziehungen und des prä-diagnostischen Verlaufs basieren auch auf Audio- und Videoaufnahmen, die uns von betroffenen Patienten und deren Familien weltweit zur Verfügung gestellt werden. Diese internationalen Kooperationen stellen uns vor das Problem technisch unterschiedlicher Aufnahmesysteme. Ein Video-Konverter würde unsere Arbeit erheblich erleichtern und den Datenverlust minimieren. RFA is a minimally invasive form to treat cancer without open surgery, by placing a needle inside the malignancy and destroying it through intensive heating. Though the advantages of this approach ale obvious, the intervention is currently hard to plan, almost impossible to monitor or assess, and therefore is not the first choice for treatment. IMPPACT will develop a physiological model of the liver and simulate the intervention's result, accounting for patient specific physiological factors. Gaps in the understanding of particular aspects of the RFA treatment will be closed by multi-scale studies on cells and animals. New findings will be evaluated microscopically and transformed into macroscopic equations. The long-established bio-heat equation will be extended to incorporate multiple scales. Validation will be performed at multiple levels. Images from ongoing patient treatment will be used to cross check validity for human physiology. Final validation will be performed at macroscopic level through visual comparison of simulation and treatment results gathered in animal studies and during patient treatment. This extensive validation together with a user-centered software design approach will guarantee suitability of the solution for clinical practice. The consortium consists of two Hospitals, three Universities, one Research Institute and one industrial SME.                 "de": "Beim kolorektalen Karzinom handelt es sich um einen der häufigsten malignen Tumore des Menschen. Eine operative Entfernung der Tumore in den Stadien I und II ist sehr effektiv, allerdings kommt es in 20-30% der Fälle zu Rezidiven. Daher ist es sehr wichtig, jene Patienten zu identifizieren, welche mit hoher Wahrscheinlichkeit ein Rezidiv entwickeln, und somit von einer Chemotherapie profitieren würden. Veränderte DNA Methylierung in Tumoren ist ein Kennzeichen von Krebs und wurde in kolorektalen Tumoren bereits umfassend untersucht. Eine veränderte Promotor-Methylierung kann auch in frei zirkulierenden Nukleinsäuren aus Serum detektiert werden, und stellt ein neue nicht-invasive Methode dar, um Hochrisikopatienten zu identifizieren.\r\nZiel dieses Projektes ist die Etablierung der High-Resolution Melting Real-Time PCR als prognostisches Werkzeug zur Risikoabschätzung von Wiederauftreten kolorektaler Tumoren bei Stadium II Patienten.",

                "en": "Human placental villous trees are covered by a two-layered epithelium-like trophoblast layer, which consists of mononucleated cytotrophoblasts and an overlying multinucleated syncytiotrophoblast. Cell turnover in the trophoblast layer includes proliferation, differentiation and subsequent fusion of cytotrophoblasts with the syncytiotrophoblast. Thus, both layers represent stages of different degree of differentiation, ranging from very early differentiated (cytotrophoblast) to terminally differentiated stages (syncytiotrophoblast). Data on comparative whole transcriptome analyses between in vitro cultured mononucleated and multinucleated primary trophoblasts exist, but have their clear limitations. Withdrawal from the in vivo microenvironment as well as in vitro culture of primary cells alters gene expression profiles, which no longer resemble the in vivo situation. To overcome these limitations we intent to microdissect these two cell fractions directly from fixed, paraffin embedded placental tissue. Total RNAs from microdissected cytotrophoblasts and syncytiotrophoblast will be subjected to microarray analysis with the aim to detect differentially expressed genes. The expectations in this project are to gain insight into the spatial expression of genes relevant for trophoblast differentiation. Furthermore, this project should deliver the basis for follow-up studies comparing placentas from healthy and pathologic pregnancies."
