Stützgewebes Ewing und CIC-DUX4 sind sich hinsichtlich ihrer Histologie sehr ähnlich. Betrachtet man jedoch molekulare Gesichtspunkte, Alter der PatientInnen und vor allem das Outcome sind diese sehr heterogen.\r\nEwing Sarkome betreffen vor allem Kinder und Jugendliche. Die 5 Jahres Überlebens Wahrscheinlichkeit beträgt ohne Metastasierung über 70% (S1 Leitlinien Ewing Sarkome des Kindes und Jugendalters, Stand 2014, Antonescu et al. beschreibt sogar 82%). Die derzeitige Behandlung erfolgt mit dem VIDE Schema (Vincristin, Ifosfamid, Doxorubicin, Etoposid).\r\nSeltener, aber bösartiger ist das CIC-DUX4 fusionierte Sarkom. Diese Erkrankung trifft PatientInnen im Median von 31 Jahren. Allerdings sind alle Altersgruppen vertreten. CIC und DUX4 sind zwei Genabschnitte, welche fusioniert sind. Aufgrund der Tatsache, dass CIC-DUX4 und Ewing morphologisch ähnlich sind, werden sie gleich therapiert. Obwohl molekulargenetisch bereits festgestellt ist, dass es sich dabei um verschiedene Entitäten handelt. Mit dem Ergebnis, dass die 5-Jahres-Überlebensrate von Patientinnen mit CIC-DUX4 Sarkom bei 43% (Antonescou et al.) liegt.\r\nZiel dieses Projekts ist es, mögliche Resistenzmechanismen der beiden Sarkomentitäten charakterisieren zu können.\r\nDabei handelt es sich um einen Zellkulturversuch.\r\nEin wesentlicher Faktor zur Resistenz gegenüber Chemotherapeutika ist die Zellmembran. Die Zellmembran besteht aus drei Hauptlipiden: Phospholipiden, Sphingolipiden und Cholesterin. Dabei soll die Lipidzusammensetzung der Zellen mittels Massenspektrometrie analysiert werden.\r\nWeiters werden die Sarkome Ultrastrukturell auf Fett Tröpfchen analysiert. Dies kann einen Hinweis auf Resistenzen geben, denn lipophile Therapeutika (wie zum Beispiel das Doxorubicin) könnten darin gefangen werden und dadurch keinen ausreichenden Wirkspiegel im Zellkern ausbilden.\r\nDie funktionale Bestätigung einer möglichen Resistenzbidldung in CIC-DUX4 Sarkomen durch Lipiddetektion mitels Massenspektrometrie und ultrastruktureller Aufklärung in Bezug auf Fett Hence, the open conformations is the critical missing piece of information to solve the TRPC3 gating puzzle. This knowledge is essential for comprehension of the molecular function of TRPC3 and definition of possible ligand binding sites to control this channel is health and, especially, disease.\r\n• Hypotheses / research questions / objectives: Here, we propose to succeed in capturing open pore structures of TRPC3 by stabilizing and synchronizing the channels at a high open probability using photochromic ligands and in addition by preserving their lipid environment during the protein preparation for cryo-EM.\r\n• Approach / methods: We will screen two expression systems for suitability to maintain TRPC3 in open conformation. We will utilize photopharmacological tools for a temporal control over TRPC3 gating during protein extraction and cryo-EM. Structural details obtained from cryo-EM will be followed by structure-guided mutagenesis and evaluated in a multifunctional live cell approach combining Ca2+ imaging and electrophysiology.\r\n• Level of originality / innovation: With our project, we will introduce a novel approach to control protein conformations during cryo-EM by photoswitchable ligands.
