                "de": "Die Pathogenese kortikaler Läsionen sowie der Zusammenhang der verschiedenen degenerativen Prozesse im ZNS bei MS sind bis heute weitgehend unklar. Bisherige Studien zeigten, dass meningeale Entzündungen eine wichtige Rolle bei der Entstehung der kortikalen Pathologie spielen können. Aggregierte Lymphozyten und Antikörper (T- und B- Zellen) infiltrieren Meningen und perivaskuläre Bereiche und führen in weiterer Folge zu einer Progression der Erkrankung. Aus dieser Erkenntnis ergibt sich die Fragestellung, ob unmittelbar periventrikuläre (d.h. liquornahe) Marklagerläsionen mit der kortikalen Pathologie in MS in Zusammenhang stehen.Um diese Fragestellung zu untersuchen, sollen in einem interdisziplinären Forschungsprojekt der Universitätsklinik für Neurologie und der klinischen Abteilung für Neuroradiologie (Universitätsklinik für Radiologie) der Medizinischen Universität Graz existierende MRT-Daten einer unizentrischen prospektiven MS-Studie (n=170) analysiert werden. Die kortikale Pathologie wird anhand der Messung der kortikalen Dicke geschätzt. Periventrikuläre Läsionen der weißen Substanz werden halbautomatisiert segmentiert. Die MR-Parameter werden quantifiziert und mittels Korrelationsanalyse wird die Hypothese überprüft, ob verminderte kortikale Dicke (sogenanntes „cortical thinning“ als Ausdruck subpialer kortikaler Läsionen) mit einer Häufung periventrikulärer Läsionen korreliert. Eine Korrelation zwischen diesen MR-Parametern wäre möglicherweise ein Hinweis dafür, dass die Läsionsprogression in MS durch spezifische Faktoren in der zerebrospinalen Flüssigkeit beschleunigt beziehungsweise vielleicht sogar initiiert wird.",

Psoriasis ist ein eigenständiger erheblicher Risikofaktor für arterielle Erkrankungen des Herz-Kreislauf-Systems, die Ursachen für den beobachteten Zusammenhang sind jedoch unklar. In diesem Projektantrag soll untersucht  werden, ob Psoriasis mit einer funktionellen Beeinträchtigung des high-density lipoproteins (HDL) assoziiert ist. Unter physiologischen Bedingungen vermittelt anti-atherogenes HDL den reversen Cholesterintransport und wirkt vaskuloprotektiv. Mittels massenspektroskopischen und biochemischen Analysen sollen Veränderungen im Proteom und in der Lipidzusammensetzung des HDL von Psoriasis Patienten im Vergleich zu gesunden Kontrollen untersucht werden. In weiterer Folge soll untersucht werden, ob eine veränderte HDL-Zusammensetzung mit einer funktionellen Beeinträchtigung des HDL-Partikels einhergeht. \r\n",
                "en": "Psoriasis, a very common skin disease, is nowadays considered as a systemic inflammatory condition. Psoriasis patients harbor an increased risk of myocardial infarction and stroke, and numerous studies indicate that psoriasis and cardiovascular disease - particularly atherosclerosis - share common pathogenic features. \r\nPatients with autoimmune diseases such as systemic lupus erythematosus and rheumatoid arthritis have decreased levels of high-density lipoprotein (HDL), suggesting that HDL may be important in preventing autoimmunity and the loss of self-tolerance. Proposed mechanisms for the atheroprotective functions of HDL include reverse cholesterol transport, reduction of oxidative stress and potent anti-inflammatory effects. On the other hand, HDL was shown to lose its-anti-atherogenic properties by post-translational modifications such as oxidation and carbamylation. There is emerging evidence suggesting that anti-atherogenic activities of HDL cannot be described by its plasma concentrations alone. For example, cholesterol efflux capacity, a functional measure of HDL, has an inverse association with the likelihood of coronary artery disease independent of HDL-cholesterol levels. Recent studies that elucidated the proteome of HDL from healthy individuals and patients with coronary artery disease or renal disease by mass spectrometry revealed that the protein cargo is a major determinant of the anti-atherogenic and anti-inflammatory properties of HDL. As qualitative alterations of HDL seem to be directly linked with increased cardiovascular complications, we hypothesize that HDL from psoriatic patients might display defective cholesterol efflux capability and anti-oxidative activities. \r\n"
