Biologische Proben und Daten werden daher auch\r\nals die Schlüsselressource für den Fortschritt in Medizin und Biotechnologie gesehen (OECD 2001).\r\nDie nachhaltige Erschließung dieser Schlüsselressourcen soll durch die pan‐europäische\r\nForschungsinfrastruktur BBMRI‐ERIC mit dem Hauptquartier in Graz/Österreich sichergestellt\r\nwerden. Therefore, biological samples are considered as the key resource for the\r\nadvancement of medicine and biotechnology (OECD 2001). The sustainable exploration of this key\r\nresource should be assured by the pan‐European research infrastructure BBMRI‐ERIC with its\r\nstatutory seat and executive management office in Graz/Austria. The scientific participation of\r\nAustria in BBMRI‐ERIC is exerted by a national node (BBMRI.AT) which should closely collaborate\r\nwith the central executive management office, and involves the Austrian public medical universities,\r\nthe Alpen Adria University Klagenfurt, the University of Veterinary Medicine Vienna, and the Life\r\nScience Governance Institute.\r\nThe work programme will Mehrere neue Arten solcher Lipidmoleküle, welche die Funktion von Krebszellen beeinflussen können, wurden in den letzten Jahren entdeckt. Unter ihnen befinden sich die sogenannten Endocannabinoide, die ähnliche Wirkung haben wie die Inhaltsstoffe von Cannabis sativa, aber im Organismus selbst gebildet werden. Diese Lipide sind Botenstoffe an Krebszellen, wo sie an Rezeptoren binden und die Funktion der Zellen beeinflussen.\r\nIm vorliegenden Projekt wollen wir einen sogenannten atypischen Cannabinoid Rezeptor mit dem Namen GPR55, der an Dickdarmkrebszellen vorkommt, untersuchen. Dieser Rezeptor besitzt als aktivierenden Botenstoff nicht nur Endocannabinoide, sondern auch ein Phospholipidmolekül, genannt LPI (Lysophophatidylinositol). LPI und GPR55 bilden zusammen eine Achse, von der man glaubt, dass sie eine treibende Rolle im Dickdarmkrebs und in entzündlichen Darmerkrankungen spielt. Diese Hypothese wird durch die Tatsache untermauert, dass GPR55 sowohl in Dickdarmkrebszellen als auch im gesunden Darm vorhanden ist und in experimentellen Modellen die Entzündung und das Krebswachstum fördert. Zur Klärung der Frage, ob diese molekulare Achse auch in Darmentzündungen und im Dickdarmkrebs des Menschen Bedeutung hat, wird der Gehalt an GPR55 Rezeptoren im Tumorgewebe von Kolonkarzinompatienten und in Darmbiopsien von Patienten mit entzündlichen Darmerkrankungen mittels geeigneten biochemischen und histologischen Methoden nachgewiesen. LPI wird im Serum mittels hochauflösender Massenspektrometrie gemessen. \r\nDie wenig erforschte Rolle von GPR55 in diesen Erkrankungen wird in diesem Projekt erstmals in humanen Proben untersucht. Es wird vermutet, dass GPR55 als Bestandteil des Endocannabinoidsystems an wichtigen Schaltstellen zur Entstehung dieser Erkrankungen sitzt. Diese Tatsache macht GPR55 zu einem wichtigen Angriffspunkt einer pharmakologischen Therapie.
