Für diese zellulären Prozesse strömt Kalzium über eine Pore, den sogenannte Ionenkanal Orai1, in die Immunzelle ein. Orai1 wirkt somit wie ein Schalter, den die Immunzelle entweder oszillatorisch oder langanhaltend öffnen kann. Mutationen in Orai1 können zu schweren Immunkrankheiten führen. In diesem Projekt möchten Dr. Rainer Schindl und seine Mitarbeiter herausfinden, wie die Genregulation der Immunzellen durch molekulare Strukturen des Kalziumtransports gesteuert wird. In Vorexperimenten konnten wir bereits mit Supercomputern eine Bindung von Kalzium an den Ionenkanal Orai1 simulieren. Unsere Hypothese ist, dass diese Kalzium Bindedomäne den Kalzium Einstrom reguliert, und für die Genregulation von entscheidender Bedeutung ist. Zur experimentellen Untersuchung wird die Kalzium Bindestruktur genetisch verändert und mit dem ursprünglichen Orai1 Ionenkanal verglichen. Sowohl der Einstrom von Kalzium in die Zelle, die Aktivierung von Genen als auch die anschließende Produktion von Proteinen werden gemessen. Diese Methoden erlauben es eine mechanistische Verbindung von molekularen Orai1 Strukturen über Kalzium Transportwege bis hin zur Produktion von Proteinen herzustellen. Die Charakterisierung der Kalzium Bindestruktur im Orai1 Kanal verspricht fundamental neue Erkenntnisse über den Kalziumtransport in die Immunzelle. This signaling process controls both the proliferation of immune cells and the defense against bacteria. To mediate these cellular processes, calcium influx occurs via special pores, termed the Orai1 ion channel. Orai1 acts like a trigger that can be oscillatory or sustained opened by an immune cell. Mutations within Orai1 can result in severe immune diseases. Within this project, Dr. Rainer Schindl and his team aim to resolve which molecular structures are required for the calcium transport into immune cells and subsequent gene regulation. In preliminary experiments, we used super computers to simulate the binding of calcium with the ion channel Orai1. We hypothesize that this calcium binding domain is crucial for gene regulation. Hence the calcium binding structure will be genetically modified and compared experimentally with the function of the original Orai1 ion channel.
