Ex-vivo organ slices derived from human surgically resected organs serve as short-term in-vitro models and alternatives (3R’s) to animal experiments. These models offer advantages over classical 2D cell culture systems and organoid models by preserving the functional properties, the cellular heterogeneity and the structural architecture of the original organ for a defined period.

The generation of PCOS from human specimens can be instrumental in addressing specific research questions across various applications, including:

Host-pathogen interaction studies (focus and experience on human respiratory viruses)
Testing of antivirals and small molecules
Drug screening, drug metabolism and toxicity

Standard histology and molecular biology methods can be applied to PCOS as read-outs, including immunohistochemical staining, gene expression analyses, and protein analyses.

Proteomics denotes the experimental analysis of proteins and proteomes, mostly determined via mass spectrometry. The output often relies first on a well thought experimental design. Secondly on a sample preparation method depending on the research question. And third: The right handling of mass spectrometry data is crucial for a reliable output. In this course you will learn the most important facts on experimental strategies and how to set up an proteomics experiment. Additionally there will be an introduction in mass spectrometry, for a better understanding what Proteomics is doing. Further there will be a hands on training on a small proteomic dataset to get even more insight into proteomic analysis.

For the hands on training the software Perseus is used. You can download it for free in advance.

The Estimand framework: An introduction

The first presentation is an introduction into the estimand framework. It first discusses why the framework is needed and was developed in an ICH guideline. And then it introduces all the elements of the framework and how they are implemented. The key elements of an estimand - the five attributes - will be discussed in detail. Specific focus will be on intercurrent events and the different strategies how to handle them. An example on the implementation of the framework in primary progressive MS will finally illustrate the framework and its usefulness.

The Estimand framework: Successes, Issues and Examples

The second presentation on the estimand framework will provide first a critical review where the industry and regulatory bodies around the world stand with respect to the implementation of the framework, what has so far worked well and what worked less well. The presentation will then focus on the two challenging areas of the framework and highlight there the issues, the choice of the intercurrent event handling strategy and the subsequent analytical approach. All the issues will then be discussed in three examples. The first one is a discussion of the treatment policy strategy, when it makes sense and when not. The second one deals with the principal stratum strategy in an example in MS. And finally, the third example deals with the implementation of the framework, when a strategy is easy to implement and when it becomes complex. All examples together highlight the challenges but also the usefulness of the estimand framework.

Sample Size Determination: Power and Minimal Detectable Difference

The first presentation introduces and discusses two different ways to justify the size of a study, one is the usual power calculation, we are all familiar with, and the other approach is based on the minimal detectable difference. The reasons for introducing the concept of the minimal detectable difference is to align statistical significance with clinical relevance. The presentation then discusses two examples. The first example deals with the size of a cardiovascular outcome study where typically a realistic effect is close to a minimal clinically relevant difference. And the second example discusses the sample size calculation of a confirmatory trial in Alzheimer's disease where different issues occur, making again the concept of a minimal detectable difference important.

Non-inferiority assessments: Overview and issues

The second presentation discusses the assessment of non-inferiority in clinical trials. It presents some of the underlying methodology (high level), issues and also provides a number of examples. Major issues will be discussed like the choice of the population, the constancy assumption and the determination of the non-inferiority limit. All these issues point basically to the overarching issue of assay sensitivity in a non-inferiority trial. For the determination of the non-inferiority limit key approaches will be introduced, a basic definition based on clinical relevance and two retained effect methods, the 95-95% method and the synthesis method. Three examples will finally illustrate the importance of all these elements.

Unser Gehirn ist ein ausgezeichneter Detektor für Muster. Wir nutzen Kategorien und Faustregeln, wenn wir wahrnehmen und entscheiden. In der Regel läuft diese Einschätzung richtig. Doch manchmal werden diese unbewusst durch kognitive Verzerrungen, den Unconscious Biases, verfälscht.

Die Folgen sind vielfältig: Wir über- oder unterschätzen die Kompetenzen von Menschen, wir präferieren Personen die uns ähnlich sind oder wir lassen uns meist unbewusst von Bias-Dynamiken auf der Gruppenebene beeinflussen.

In diesem Workshop zeigt der Bias-Experte Roland Engel auf, wie Unconscious Biases entstehen und welche Arten speziell im universitären Alltag die fachliche Entscheidungsfindung trüben können. Im Praxistransfer werden anschließenden jene Umgangsstrategien diskutiert, die sich in den letzten Jahren vor allem im Universitätsbetrieb bewährt haben.

Student*innen der Medizinischen Universität Graz werden am Samstag, dem 19.10.2024 im Rahmen der „Drück Mich“-Initiative in ganz Graz als fliegende Teams unterwegs sein, um Passant*innen in kürzester Zeit zu demonstrieren, wie einfach die Wiederbelebung bei einem unerwarteten Herzstillstand ist.

Der unerwartete Herzstillstand – keine Seltenheit!

Jeder Zehnte in Österreich erleidet im Laufe seines Lebens einen unerwarteten Herzstillstand. Treffen kann es dabei jede und jeden, jederzeit – ungeachtet des Alters. Momentan überleben nur etwa zehn Prozent ein solches Ereignis. Die Initiative „Drück mich!“ hat es sich zum Ziel gesetzt, möglichst vielen Menschen die Wiederbelebung beizubringen. Dadurch könnten jedes Jahr zusätzlich 1.000 Leben in Österreich gerettet werden.

Slurm is an open-source, fault-tolerant, and highly scalable cluster management and job scheduling system for large and small Linux clusters.

It provides three key functions:

1) allocating exclusive and/or non-exclusive access to resources (nodes) to users for some duration of time so they can perform work,

2) providing a framework for starting, executing, and monitoring work (typically a parallel job such as MPI) on a set of allocated nodes, and

3) arbitrating contention for resources by managing a queue of pending jobs.

This course will go over the basic principles of using the Slurm Workload Manager on the MedBioNode cluster. There will be some short hands-on exercises, so attendees should know how to work in Linux and log on the cluster as a prerequisites.

Most tasks of bioinformatics are processed using the Linux operating system (OS). Therefore, familiarity with and understanding of basic Linux command lines is essential for bioinformatics analysis. This course will provide you an introduction to the Linux OS and its basic command line tools. Material will cover logging into remote machines, filesystem organization and file manipulation. During the course participants will also learn how to use and submit jobs with SLURM on the cluster.

Upon successful completion of this course, you will be familiar with:

Basic Linux commands (including grep, sort, find, etc.)Running tools over SLURM (cluster management and job scheduling system)

Kari Alitalo, from the Faculty of Medicine of the University of Helsinki and Director of the Wihuri Research Institute, will talk about “Translational insights into endothelial growth factors”:

Vascular endothelial growth factors (VEGFs) are involved in orchestrating the development, maintenance and regeneration of the blood and lymphatic vessels. However, despite significant progress in mechanistic understanding of the function of VEGFs in various human disease processes, there has been limited translation of this knowledge to disease therapeutics. In my talk, I will review some of our attempts to make headway towards better understanding and proof-of-principle clinical translation of the VEGFs.
