However, there is still little known on how LAMB3 behaves after implantation of the embryo into the endometrium. This master thesis aims to find out how the expression of LAMB3 changes with the invasion of the extravillous trophoblasts (EVTs). Therefore, we established a double immunofluorescence staining protocol and detected LAMB3 and EVTs together on a formalin fixed, paraffin embedded (FFPE) placental serial section using an anti-LAMB3 and anti-HLA-G antibody. The extravillous trophoblasts invade from the fetal vili into the maternal decidua. The samples therefore contain invaded and non-invaded tissue. The expression of LAMB3 in both areas was analyzed using image-based quantification and the CellProfiler software. In two out of three placental tissues the LAMB3 expression was increased in the invaded area compared to the non-invaded area. At the same time, we also attempted the first preliminary experiments for organoid cultivation. Organoids resemble the original tissue, are genetically stable and are well suited as a 3D culture system for in vitro research. The digestion of endometrium and placenta samples was optimized. Cytospins and immunohistochemistry were used to check the cell yield. Glandular epithelial fragments were isolated. This master thesis provides the basis for further LAMB3 research with a larger cohort of placenta samples.
