A major barrier to curative AML treatment is the persistence of leukemic stem cells (LSCs), a subpopulation resistant to standard therapy (1). Thus, there is an urgent need for innovative therapeutic strategies that specifically target this resilient cell population.Increasing evidence links inflammatory signaling to both healthy and malignant hematopoiesis, influencing hematopoietic stem cell (HSC) function and leukemogenesis. Within the inflammatory network, tumor necrosis factor receptor (TNFR)-mediated signaling is a key regulator of inflammation and cell death and has been shown to be crucial for maintaining healthy hematopoiesis (2). The receptor proximal adaptor molecule TNF receptor-associated factor 2 (TRAF2) is a key modulator of TNFR signaling, directing the activation of pro-survival NF-kB pathway while simultaneously controlling cell death responses. However, the precise role of TNFR signaling in the context of AML development remains poorly understood (3). Previous work from our laboratory demonstrated that activation of TNFR mediated inflammatory signaling can induce cell death and differentiation in LSCs while sparing healthy hematopoietic progenitors, highlighting a promising therapeutic avenue for selectively targeting and eliminating leukemic cells (4).
