            "abstract": "Herz-Kreislauf-Erkrankungen sind weltweit die häufigste Ursache für Morbidität und Mortalität, die frühzeitige Identifikation von Risikokonstellation stellt eine Herausforderung dar. Die Gefäßfunktion spielt eine zentrale Rolle in der kardiovaskulären Gesundheit und wird zunehmend im Kontext nicht-invasiver funktioneller Diagnostik untersucht. Die Herzratenvariabilität (HRV) gilt als sensibler Marker der autonomen Regulation und spiegelt das Zusammenspiel von Sympathikus und Parasympathikus wider. Ziel dieser Arbeit war es, den Zusammenhang zwischen HRV-Parametern und Gefäßfunktion zu analysieren sowie zu evaluieren, ob eine mechanische Stimulation mittels „Wobbler“ messbare Veränderungen im autonomen Nervensystem und der Gefäßreaktion hervorruft.",

            "title": "Retrospektive Auswertung der Gerichtsverfahren bei Verdacht auf schwere Kindesmisshandlung mit und ohne Todesfolge vor Vollendung des 1. Lebensjahres in Österreich",
            "abstract": "Einleitung: Kindesmisshandlung im Säuglingsalter stellt ein gravierendes medizinisches, gesellschaftliches und juristisches Problem dar. Säuglinge sind aufgrund ihres Alters und ihrer völligen Abhängigkeit besonders vulnerabel gegenüber Gewalt und Vernachlässigung. Im Vergleich zu allgemeinen Strafverfahren (bei Erwachsenen ≥14 Jahre) zeigte sich eine signifikant niedrigere Verurteilungsrate bei Säuglingen (35,9% vs. 46,5%, Chi-Quadrat-Test p<0,001). Ziel dieser Arbeit war es, anhand österreichischer Daten des Justizministeriums, das Verhältnis zwischen Anklagen und Verurteilungen bei Misshandlungsfällen mit und ohne Todesfolge im ersten Lebensjahr zu analysieren. Zudem wurden Deliktarten, regionale Verteilung und gerichtliche Erledigungsformen untersucht und mit Daten erwachsener Opfer verglichen, um Muster und mögliche Schwachstellen in der Aufarbeitung solcher Fälle aufzuzeigen.",

            "title": "The Role of Succinate in Pulmonary Fibrosis",
            "abstract": "Idiopathic pulmonary fibrosis or IPF is a chronic and progressive interstitial lung disease associated with a poor post-diagnosis survival rate of 3-5 years. Characterized by repetitive scarring of lung tissue, loss of alveolar integrity and compromised gas exchange, IPF has limited treatment options with patients ultimately requiring lung transplantation. While current therapeutics slow disease progression and lung function decline, they do not halt or reverse the established disease. Thus, there is a pressing need to identify novel curative therapeutic strategies. Metabolomic studies have revealed that several metabolic pathways including glucose metabolism, amino acid metabolism and lipid metabolism are altered in IPF patients. Importantly, studies from the last decade have also highlighted dysregulation of the tricarboxylic acid (TCA) cycle or the Krebs cycle which is a central hub to several metabolic pathways.",

            "abstract": "Eine starke Gesundheitskompetenz hilft dabei, Entscheidungen zu treffen, die zu einer Verbesserung oder Aufrechterhaltung der Lebensqualität führen. Gesundheitsinformationen (GI) sollen die Gesundheitskompetenz der Bevölkerung steigern. In den allgemeinmedizinischen Praxen in der Steiermark weisen jedoch die meisten verfügbaren GI erhebliche Mängel in Bezug auf inhaltliche Qualität, strukturelle Aufarbeitung sowie eindeutige Identifikationsmerkmale auf, wodurch sie ihren Zweck nicht erfüllen. Aus diesem Grund wurde das Projekt „Evidenzbasierte Information zur Unterstützung von gesundheitskompetenten Entscheidungen“ (EVI) entwickelt. Da das Thema unkomplizierte Harnwegsinfektion (uHWI) bislang nicht in die EVI-Initiative integriert wurde und sich laut einer Befragung 5,06% der Allgemeinmediziner*innen qualitativ hochwertige GI dazu wünschen, ist eine Sichtung und Evaluierung der vorhandenen Materialien nötig. Ziel dieser Arbeit war es, bereits vorhandene GI zum Thema uHWI zu evaluieren, um inhaltlich leitliniengerechte GI für die Aufnahme in die EVI-Initiative auszuwählen.",

            "abstract": "Fasting activates a coordinated systemic response involving key metabolic tissues, like adipose tissue (AT) and liver. At the onset of fasting, AT rapidly mobilizes stored lipids, while the liver initiates glycogenolysis followed by gluconeogenesis and ketogenesis to maintain long-term energy homeostasis. Key players in these adaptations are transcription factors (TFs) that bind to promoter and enhancer regions to coordinate cellular transcriptional programs. Although individual fasting-responsive regulators have been described, their time-resolved coordinated activity remains not fully understood. Transcriptionally active TF-bound enhancers generate short-lived enhancer RNAs (eRNAs). Using quantitative precision run-on sequencing (qPRO-seq) we generated the first nascent transcription-based atlas of fasting-regulated genes and enhancers in mouse liver and AT.",

            "abstract": "Hintergrund: Neuroendokrine Tumoren der Lunge (pulmonale NET) bilden eine heterogene Entität mit variabler biologischer Aktivität und Prognose. Die präzise histopathologische Klassifikation ist essenziell für die Wahl geeigneter therapeutischer Strategien und die Abschätzung des klinischen Verlaufs. Während der Proliferationsmarker Ki-67 in der Diagnostik gastroenteropankreatischer NETs etabliert ist, wird seine diagnostische und prognostische Relevanz bei pulmonalen NET bislang zurückhaltender behandelt. In der aktuellen WHO-Klassifikation 2021 wird der Ki-67 Proliferationsindex als wünschenswertes Kriterium genannt. Ziel der vorliegenden Arbeit war es, im Rahmen eines systematischen Literaturreviews den aktuellen Kenntnisstand zur Bedeutung von Ki-67 bei neuroendokrinen\nLungentumoren zusammenzufassen und dessen potenziellen Stellenwert für die zukünftige Integration in die standardisierte Diagnostik zu evaluieren. Analysiert wurden internationale Leitlinien, technische Aspekte der Ki-67-Bestimmung, deren Reproduzierbarkeit sowie der prognostische und differentialdiagnostische Nutzen des Markers.",

            "abstract": "Einleitung: Muskelverletzungen zählen im Profifußball zu den häufigsten Ursachen für Ausfallzeiten. Die Magnetresonanztomographie hat sich dabei als zentrales diagnostischen Verfahren etabliert. Obwohl derzeit eine Vielzahl an MRT-Klassifikationssysteme vorliegen, besteht weiterhin kein Konsens über deren prognostische Relevanz. Auf dieser Grundlage wurde das Bildmaterial des Studienkollektivs mit den gebräuchlichsten Klassifikationssystemen abgeglichen und den klinischen Informationen gegenübergestellt. Ziel dieser Arbeit war es, die prognostische Wertigkeit der MRT-Diagnostik bei Muskelverletzungen und ihre Aussagekraft bezüglich der Rekonvaleszenz zu untersuchen.",

            "title": "Maternal Obesity in Early Human Pregnancy: Placental DNA Integrity and Stress Response",
            "abstract": "In the first trimester (FT) of human pregnancy, the intrauterine environment directly affects development of the rapidly growing placenta. Maternal obesity - a frequent manifestation of metabolic derangements - is associated with metabolic complications, systemic low-grade inflammation and genotoxicity. Considering that the developing placenta is exposed to obesity-associated changes in the early intrauterine environment, we hypothesize that maternal obesity affects DNA integrity that triggers DNA damage responses (ATM/ATR signaling) and affects placental stress response with consequences for trophoblast turnover in early human pregnancy, respectively. The present study aimed to determine FT placental DNA damage, telomere shortening and subsequent effects on DNA damage repair and cell cycle regulation. Furthermore, we aimed to characterize FT placental stress status and final effects on trophoblast proliferation and apoptosis. \r\nFor this purpose, placental tissue (week 4-12 p.m.) was obtained from non-smoking (self-report and serum cotinine measurement) women terminating pregnancy for psycho-social reasons. DNA damage and oxygen-induced modification in total tissue were determined by COMET assay. We quantified DNA damage (γH2AX) and oxidative DNA modification (8-OHdG), proliferation (Ki67) and apoptosis (TUNEL, caspase cleaved cytokeratin 18) in villous cytotrophoblasts (CTB) using a semi-quantitative in situ analysis based on immunofluorescence triple-staining. Telomere length in total placental tissue and laser-capture microdissected trophoblast was quantified by RT-qPCR and confirmed by quantitative fluorescence in situ hybridization (Q-FISH). Pre-selected placental stress markers, cytokines, DNA damage repair and cell cycle related genes were assessed using a PCR-gene panel and Nanostring assay, while proteins were quantified by a cell cycle-specific protein array or immunoblotting. Data was analyzed using a multivariate linear regression model with adjustments for gestational age and tissue processing time.\r\nThe key findings of this study were: \r\ni. Maternal obesity in early pregnancy was not associated with oxidative DNA modifications (8-OHdG) in total tissue and CTB. Other indicators of oxidative stress such as products of lipid peroxidation and protein nitration as well as the classical stress marker HSP70 were unaltered by maternal obesity in early pregnancy. Furthermore, gene expression of placental antioxidants (MGST3, SOD3, GSTP1, GLRX, TXNRD1) and heme oxygenase 1 (HO-1) was lower in obese pregnant women, while placental Nrf2 expression and HO-1 protein levels were not altered in samples from obese pregnant women.\r\nii. ii.Maternal obesity affected placental inflammatory cytokine expression (IL1A, IL6) and stress signaling genes (ASK2, MAPK11, MAPK12, MAPK13, JNK1) in FT placentas.\r\niii. iii.Maternal obesity induced placental DNA damage specifically in CTBs. CTB telomeres in situ were shorter in maternal obesity. Shortening was associated with high C-peptide levels, low insulin sensitivity or high BMI. Despite higher ATM/ATR protein levels and increased substrate phosphorylation in FT placentas of obese pregnant women, selected downstream proteins remained unaffected, while central DNA damage repair transducers (HDAC1, HDAC8, PARP1, FEN1, ku70, BLM) were downregulated in FT placentas from obese pregnant women. This dysregulated damage repair was also accompanied by an induction of cell cycle arrest. \r\niv. iv.Finally, we found decreased CTB proliferation and an increase in apoptosis in FT placentas from obese pregnant women, while senescence was unaltered. \r\nAltogether, these results argue for absence of oxidative stress in first trimester placentas of obese women. They also suggest that maternal obesity in the first trimester affects CTB turnover and therefore alter placental development, potentially ensuing adverse consequences for maternal and fetal health.",
