Its function is highly versatile and complex. β-Catenin is the key nuclear effector of a major pathway for numerous developmental and homeostatic processes, the Wnt pathway. The importance of this pathway shows upon the pathways dysfunction that results in severe diseases, such as multiple types of cancer, skeletal defects and birth defects such as spina bifida. β-Catenin acts not only as a Coactivator of the Wnt pathway, but also in a Wnt-independent manner, by interacting with different other transcription factors. This crosstalk with other pathways as well as the increasing number of studies to understand the protein’s complex function, make β-Catenin a very requested protein to further investigate.\r\n\r\nDiving into the β-Catenin-HIF-1α interaction as well as the structural resolving of the interacting TAD would deliver more mechanistic understanding and approaches for drug development, especially in the case of cancer. This thesis, therefore, aims to investigate the dynamic and structural properties of the HIF-1α fragment [478- 580 aa]. Additionally, it intends to elucidate the β-Catenin-HIF-1α interaction by capturing the variation in residue involvement, to validate ArmN (N-terminal part of the armadillo repeat domain) as the interaction location within the β-Catenin protein, and to estimate the binding affinity.\r\n\r\nThis thesis derives from a machine learning model created by Yukti Khana and extends the preceding investigations, that tested the binding interactions β-Catenin has with p53, FOXO1, FOXO3, FOXO4, FOXO6, TCF7, FOXM1, FOXP2, and LEF1. (section 1.2.1 The machine learning model) The experimental part involved the data collection and analysis of multiple 2D and 3D NMR spectra along with various experimental investigations. Furthermore, fluorescence polarization assays were conducted.\r\n\r\nOur data show that the HIF-1α fragment [478- 580 aa], is mainly intrinsically disordered but still has tendencies for β-strands and α-helices. The predicted TAD that lies within the N-TAD shows clear binding in HSQC (free vs bound). Due to the interaction pattern, the involvement of residues could be identified; the similar interaction pattern tested with the full length β-Catenin and N-terminal part of the armadillo repeat domain suggests that the ArmN region is involved in the interaction within β-Catenin.\r\n\r\nSince the outcome of the Fluorescence Polarization Assay that was tested within the β-CateninΔIDR up to 100 µM showed no plateau, the saturation is believed to be reached at a higher β-Catenin concentration, therefore indicating a high dissociation constant and accordingly a weak interaction.", "authors": [ "Kazman, AS" ], "year": 2026, "source": " [ Diplomarbeit/Master Thesis (UNI) ] TU Graz; 2026. pp.103.
