The proteins' involvement in numerous developmental stages is demonstrated by the significant pleiotropic abnormalities observed in HIPP knockouts. Thus far, it has been demonstrated that HIPPs contribute to long-distance plant mobility, regulate cytokinin responses, and help plants adapt to biotic and abiotic stress. HIPP deletion mutants had a high number of differentially expressed genes (DEGs), according to RNA-sequencing data, confirming their essential biological role. According to the functional characterisation of DEGs, HIPPs are involved in the transmission of hormone signals, particularly auxin signalling. It was discovered by yeast-two hybrid screening that HIPPs interact with a wide variety of proteins, which is in line with their inherently disordered character. It's interesting to note that members of different transcription factor families made up around half of the discovered proteins and were significantly overrepresented among the HIPP interactors. We made the decision to thoroughly investigate the connections between HIPP and ARF19, a member of the auxin response factor family, considering the RNA-sequencing results showing the function of HIPPs in controlling the plant hormone auxin signalling, as well as several auxin-related abnormalities seen in the HIPP mutants.\r\n\r\nIn this thesis, the molecular interaction of ARF19 and HIPP32 is investigated by applying NMR spectroscopy. The study suggests two possible interaction mechanisms, binding of the ARF19 PB1 to the HIPP32 HMA and binding of the ARF19 DBD to the HIPP32 IDR. This HMA-PB1 interaction can severely influence the cytoplasmic condensate formation of ARF19, which is crucial in the regulation to auxin induced gene expression. Interaction between the ARF19 DBD and the HIPP32 HMA can have several effects, among other things, it influences the ability of DNA binding of the DBD.\r\n\r\nTo validate the interaction mechanisms identified by NMR and to assess their biological relevance, complementary biophysical techniques such as SPR and smFRET should be applied.
