Um dieses Hindernis zu überwinden, nutzten wir einen verbreiteten Trick in der FACS Analyse, um Zellen aus der „Side Population“ zu isolieren und zu sortieren, in welcher Stammzellen und Vorläuferzellen häufiger sind als in anderen Populationen. Der Trick, um die „Side Population“ in der FACS Analyse zu identifizieren, ist das Auspumpen von Fluoreszenzfarbstoffen, wie Hoechst 33342 oder Vybrant DyeCycle Violet, zu stoppen.\r\nMassen an Zellen und Einzelzellen wurden mit FACS isoliert und mit qPCR analysiert. Um das zu tun, nutzten wir die Strategie der „global preamplification“, um die Menge der generierten cDNA, vor dem Testen der Genexpression mit qPCR, zu vervielfältigen.\r\nDas primäre Ziel dieser Studie war die Etablierung des „Side Population“ Protokolls für die FACS Analyse und das Protokoll der „global preamplification“ von der Gruppe um Thomas Kroneis und Anders Ståhlberg et al. Wir etablierten das FACS Protokoll mit MCF-7 Zellen und das „global preamplification“ Protokoll mit MCF-7 und ACH-3P Zellen.
