Their dysregulation often contributes to disease mechanisms in neurodegeneration and cancer. This thesis integrates nuclear magnetic resonance spectroscopy (NMR) metabolomics, NMR of proteins and de novo peptide design to address four research aims. First study explores region specific metabolic differences linked to altered arginine methylation in distinct subtypes of frontotemporal dementia with TDP-43 pathology and Alzheimer’s disease. The second part of the thesis dissects in vitro the interaction between the transcription factor FOXM1 and Wnt signalling component β-catenin, revealing a binding mechanism in which β-catenin engages multiple sites within FOXM1. Finally, solvent paramagnetic relaxation enhancement measurements were corrected by applying a theoretical model to minimize solvent-exchange driven errors, improving analysis for highly solvent-exposed disordered proteins. Diese chronischen Stressoren begünstigen das Burnout-Syndrom, das erhebliche Konsequenzen für betroffene Ärzt*innen mit sich bringt: Depression, Schlafstörungen und Suizidalität. Zugleich beeinträchtigt Burnout die Patient*innenversorgung durch mehr Behandlungsfehler, gesteigerte Unzufriedenheit und längere Krankenhausaufenthalte. Zusätzlich belasten hohe Ausfallkosten das Gesundheitssystem. Achtsamkeitsbasierte Interventionen (ABI) finden seit den 1990er-Jahren zunehmend Einsatz im beruflichen Kontext, sowohl präventiv als auch therapeutisch. Metaanalysen und systematische Reviews legen nahe, dass Achtsamkeitsbasierte Interventionen Stress und Burnout senken und Wohlbefinden sowie Leistungsfähigkeit fördern. Die steigende Prävalenz dieser Erkrankung erhöht den Bedarf an evidenzbasierten Leitlinien zur Unterstützung von Gesundheitsberufen bei der Betreuung von Personen mit Adipositas. Die bestehenden Leitlinien zu Ernährungs- und Bewegungsinterventionen für erwachsene Personen mit Adipositas sind teils widersprüchlich und von variabler Qualität. Ziel: Das Hauptziel dieser Arbeit ist es, einen Überblick über Handlungsempfehlungen für Ernährungs- und Bewegungsinterventionen bei erwachsenen Personen mit Adipositas auf der Grundlage aktueller internationaler Leitlinien zu geben. Diese Empfehlungen sollen Pflegepersonen und andere Gesundheitsberufe dabei unterstützen, eine evidenzbasierte Praxis umzusetzen und die Versorgung von Personen mit Adipositas zu verbessern. Methoden: Es wurde eine strategische Literaturrecherche in Datenbanken wie PubMed und Embase sowie auf den Websites relevanter medizinischer und pflegerischer Organisationen durchgeführt. Die Qualität jener Leitlinien, welche die Einschlusskriterien erfüllten, wurde mit dem AGREE-II-Instrument bewertet. Die extrahierten Ernährungs- und Bewegungsempfehlungen aus den Leitlinien wurden systematisch zusammengefasst und gegenübergestellt. Ergebnisse: Die Literaturrecherche ergab, dass nur wenige evidenzbasierte Leitlinien existieren, die spezifische Empfehlungen zu Ernährungs- und Bewegungsinterventionen für erwachsene Personen mit Adipositas enthalten. Die Empfehlungen sind teils widersprüchlich und die Qualität variiert. Die extrahierten Empfehlungen umfassen verschiedene Aspekte wie die Art der Ernährung, die Intensität und Art der körperlichen Aktivität und den Umgang mit Komorbiditäten. Es wird etwa zu einem individuellen Ansatz in der Ernährungstherapie, zu einer Reduktion der Kalorienzufuhr, zur Portionskontrolle, sowie zur Einhaltung des mediterranen Ernährungsmodells geraten. Die Bewegungsempfehlungen beinhalten unter anderem körperliche Bewegung allgemein, körperliche Aktivität bis zu 250 und auch über 250 Minuten/Woche, sowie intensives Bewegungstraining. Schlussfolgerung: Pflegepersonen haben in ihrer täglichen Praxis häufig wenig Zeit und Ressourcen, um sich ausführlich mit evidenz-basierten Leitlinien auseinander zu setzen. Die Entwicklung evidenzbasierter Handlungsempfehlungen für Ernährungs- und Bewegungsinterventionen bei erwachsenen Personen mit Adipositas ist daher eine wichtige Maßnahme zur Verbesserung der Pflegepraxis. Pflegepersonen können von klaren, praxisnahen Empfehlungen profitieren und damit leichter evidenzbasierte Empfehlungen in die Pflegepraxis implementieren.
