As one of the primary hallmarks of aging, cellular senescence represents a suitable starting point for intervening in the emergence of age-related diseases and enhancing longevity. Senescence is characterized by cellular hypertrophy and permanent cell cycle arrest while still being metabolically active. The key hubs of metabolism are mitochondria, the main place of intracellular ATP synthesis. The energy production is strongly influenced by Ca2+ homeostasis due to the Ca2+-dependence of dehydrogenases of the tricarboxylic acid cycle, which provide the electron donors for later ATP synthesis via oxidative phosphorylation. Thyroid hormones, especially the biologically active form 3,5,3'-L-triiodothyonine (T3), are known to be master regulators of cellular and organismal metabolism.\r\n\r\nHence, we aimed to uncover the impact of T3 on the mitochondrial function in a cellular aging model based on oxidative stress-induced senescence. Therefore, we utilized tert-butyl hydroperoxide (tBHP)-treated fibroblasts to investigate the impact of T3 on expression levels of senescence- and mitochondrial Ca2+-related genes by qRT-PCR, performed fluorescence microscopy to measure mitochondrial ATP synthesis and cellular Ca2+ homeostasis, and checked senescence by the SA-β-galactosidase staining.\r\n\r\nWe found that tBHP-induced senescence is linked to increased cytosolic Ca2+ levels, a drop in the mitochondrial membrane potential, and alterations in the mRNA expression levels of Ca2+ handling proteins and senescence markers. Notably, T3 treatment for 3 h showed a tendency to counteract most of these effects. Resveratrol, a naturally occurring polyphenol, has demonstrated anti-cancer potential by inducing apoptosis through inhibition of ATP synthase and subsequent enhancement of mitochondrial Ca2+ uptake. However, its clinical application is hindered by poor bioavailability and rapid metabolism. To address these limitations, novel analogs of resveratrol were developed and investigated for their ability to selectively target cancer cells. In this study, we evaluated the potential anti-cancer properties of these resveratrol analogs by assessing their effects on mitochondrial ATP levels, the activity of the sarco/endoplasmic reticulum Ca2+ ATPase SERCA, and the Ca2+ homeostasis in HeLa cells. The compounds were further tested for a potential specific impact on proliferation of cancer cells by comparing their effect on breast cancer cells MCF7 and the non-cancerous hTERT-HME1 cell line. Using live-cell imaging, we demonstrated that three novel compounds effectively inhibited mitochondrial ATP production, leading to energy stress, disruption of Ca2+ homeostasis, and subsequent cell death in cancer cells. Thereby, we also deciphered specific differences between the individual compounds regarding the degree of impact on cancerous and non-cancerous cells. Mitochondria, the powerhouses of the cell and initiators of programmed cell death, play a crucial role in energy production and the aging process. Thyroid hormones, key regulators of metabolism, profoundly impact mitochondrial function. Our previous work demonstrated that the biologically active triiodothyronine (T3) enhances mitochondrial metabolism in cancer cells by upregulating mitochondrial calcium (Ca2+) handling proteins, which in turn boost the activity of tricarboxylic adic cycle dehydrogenases and overall mitochondrial metabolism. Given that subclinical hypothyroidism is associated with slower metabolism, reduced oxidative stress, and prolonged lifespan, we hypothesize that T3 induces mitochondrial-specific signaling cascades during aging. To test this hypothesis, we induced premature senescence in HFF-1 fibroblasts using tert-butyl hydroperoxide (tBHP) and utilized this cellular aging model to investigate the impact of T3 on young and senescent cells. We measured mitochondrial nicotinamide adenine dinucleotide (NADH) index, mitochondrial Ca2+, and adenosine triphosphate (ATP) homeostasis, as well as cytosolic and mitochondrial reactive oxygen species (ROS) levels using organelle-targeted biosensors. Additionally, we analyzed the expression levels of genes related to senescence, oxidative stress defense, and mitochondrial Ca2+ handling via quantitative real-time PCR. We found that oxidative-stress induced senescence increases the colocalisation between mitochondria and the endoplasmic reticulum, boosts stress in the endoplasmic reticulum, and increases cytosolic Ca2+ and ROS levels. Interestingly, T3 seems to counteract these observed changes.
