Der letzte Schritt des Signalwegs von Glukosemessung bis Insulinausschüttung ist ein Einstrom von Ca2+Ionen aus dem Extrazellularraum. Auch die Proteine STIM1 und Orai1 führen über den Prozess des store operated calcium entry (SOCE) zu einem Einstrom dieser Ionen. Das Ziel dieser Arbeit ist es, herauszufinden, ob ein Zusammenhang zwischen dem speicherregulierten Kalziumeinstrom und der Insulinsekretion existiert. Dazu wurden eine Kultur von Pankreaskarzinomzellen aus Ratten (INS-1) gewählt und diese u.a. mit fluoreszenzmikroskopischen Techniken charakterisiert. Es wurden diverse morphologische Änderungen der Zellen gezeigt. Ein neues Verfahren diente der Messung der glukoseabhängigen ATP-Produktion in Mitochondrien mittels FRET (Förster Resonance Energy Transfer). Weiterhin wurden molekularbiologische Messungen zur Bestimmung der abgegebenen Insulinmenge angewandt und der Mercodia Insulin ELISA für die Messungen in kultivierten Zellen adaptiert. Ferner wurden zellbiologisch die Expressionslevel der Proteine mit Überexpression und Knockdown von STIM1 und Orai1 verändert. Diverse Methoden zur Normalisierung der Insulinproduktion wurden verglichen, wobei sich die durchschnittliche Zelldichte am besten bewährte.
