Trotz der Erfolge in der chirurgischen und medikamentösen Behandlung ist die Mortalität noch immer sehr hoch. Die Mikroarrytechnologie stellt eine neue High-Throughput-Methode dar, mit der die simultane Analyse der Expression von tausenden Genen möglich ist. Mit Hilfe der Genexpressionsprofile wird es möglich sein, einen Einblick in die funktionellen Veränderungen und dynamischen Vorgänge bei Karzinomen zu bekommen.\r\n\r\nGenexpressionsprofile müssten daher besonders gut geeignet sein um komplexe Veränderungen wie die Tumor-Stromainteraktion zu analysieren. Bisher wurde jedoch noch nicht gezeigt, dass aus den komplexen Daten eines Genexpressionsprofils, das deregulierete Gene des Tumors und Stromas umfasst, Rückschlüssen auf die Stromareaktion gezogen werden können. Wir haben daher die Expression von Genen, die spezifisch nur im Tumorstroma vorkommen, mit der Expressionshöhe der korrespondierenden Genprodukte mittels immuhistochemischer Analyse verglichen.\r\n\r\nEs zeigte sich, dass mittels der Zellmarker CD3 und CD11b sowie dem Gefäßmarker CD31 auch auf RNA-Ebene eine Aussage über die Zell-bzw. Gefäßdichte im Tumorstroma möglich ist. Die Mikroarraydaten von CD4, CD22 und vWF lassen keine validen Rückschlüsse auf die Proteinexpression zu. ICAM-1 eignet sich auf Basis der RNA-Daten nicht als Marker der Angiogenese, das RNA-Level korreliert aber mit der Menge an ECM-gebundenem sICAM-1, einem Prognosefaktor und Parameter der Antitumorimmunität.\r\n\r\nWeiters konnten wir zeigen, dass unter den am stärksten deregulierten Genen im Kolonkarzinom v.a. Stromagene zu finden sind, die funktionell einen sehr gewichtigen Einfluss auf Wachstumsverhalten und Malignitätspotential des Tumors haben. Mit der Klusteranalyse haben wir Untergruppen von Genen und Tumoren identifiziert, die zum überwiegenden Teil funktionell eng miteinander verknüpft sind bzw. sich biologisch weitgehend gleich verhalten.\r\n\r\nDie Kluster zeigten die enge Beziehung zwischen IL-8 und den Proteasen uPA, uPAR und PAI-1, zeigten die gleichartige Expression von Angiogenesefaktoren bFGF, HGF, PDGF-2, CD14 und TF, sowie Homologien der Expression zwischen Angiogenesefaktoren und Proteasen: Mac-25, SPARC, Stromelysin-3, UPA, PAI-1.\r\n", "authors": [ "Asslaber, MA" ], "year": 2003, "source": "[ Dissertation ] Karl-Franzens-Universität, Graz.; 2003. pp.101.
