Beide Mutationen (N88S und S90L) zerstören die N-Glykosylierungsstelle des Proteins Seipin. Dies führt zu einer Protein Aggregation im Edoplasmatischen Retikulum (ER) (Windpassinger et al.). \r\nIn dieser Arbeit wurden zelluläre Vorgänge, die zur Neurodegeneration im Silver Syndrom führen genauer untersucht. Dafür wurden AtT20 (murine pituitary tumour cells) und HUVEC (human umbilical vein endothelial cells) Zelllinien mit Seipin Wildtype und Seiping Mutanten Konstrukten transient transfiziert und ER Stressmarker untersucht. Die Ergebnisse zeigten eine klare Erhöhung des zentral ER Stress Proteins GRP78. Des Weiteren wurde die Calcium Homöostase untersucht, wodurch eine erhöhte Aktivität der SERCA (sarco(endo)plasmic reticulum CA2+ ATPase) Pumpen aufgrund von Seiping Mutanten Expression nachgewiesen wurde. Eine erhöhte SERCA Aktivität während ER Stress wurde bereits von Caspersen und Hojmann beschrieben. Resultate aus der vorliegenden Arbeit tragen enorm dazu bei, dass die SERCA Pumpen ER indizierbare Pumpen sind. Resultate dieser Arbeit zeigen, dass Seipin Mutanten Expression zur ER Stress in Zellkulturen führt. In erster Linie stellt ER Stress eine Maßnahme dar, die die Zelle schützen und den ursprünglichen Zustand wieder herstellen soll. Offenbar ist der ER Stress jedoch zu enorm für die Zelle, was zur Einleitung der Apoptose führt, welche mittels AnnexinV Färbungen, Caspase Färbungen, Caspase Hemmung oder durch kernmorphologische Veränderungen bestätigt werden konnte. Diese Forschungsergebnisse tragen zur Aufklärung der Neurodegeneration im Silver Syndrom bei. Der Ausbruch der Erkrankung liegt meist Ende der zweiten, Anfang der dritten Lebensdekade. In diesem Zeitraum kommt es in den Nervenzellen zu einer langsamen Akkumulation des falsch gefalteten Proteins und ab einem gewissen Zeitpunkt zum ER Stress.Ist der ER Stress für die Zelle nicht mehr zu bewältigen, wird das Programm der Apoptose eingeleitet und somit kommt es zur Neurodegeneration, welche sich in einer Atrophie des kleinen Handmuskels äußert. Um weitere Details betreffend der Proteinfunktion zu erhalten, wurde ein yeast two-hybrid screen durchgeführt, um Interaktionspartner von Seipin zu finden. Ergebnisse zeigen Interaktionen mit den Protein NDRG4, NETO2 und BAZ2b.
